CD99 is the product of the MIC2 gene, a glycoprotein with a molecular weight of 32kD, positively expressed in immature T lymphocytes (including thymic cortical cells), islet cells, bladder epithelium, endothelial cells, testicular sustentacular cells, and ovarian granulosa cells.
Its main application is in the diagnosis of Ewing's sarcoma and primitive neuroectodermal tumors.
CD99 is positive in B and T lymphoblastic lymphoma, acute lymphoblastic leukemia, and it also aids in the diagnosis of thymoma.
Tumors such as small cell lung cancer (Merkel cell carcinoma CD99-), islet cell tumor, carcinoid, nephroblastoma, meningioma, atypical fibroxanthoma (85%), rhabdomyosarcoma, malignant fibrous histiocytoma (35%), pilomatrixoma, giant cell vascular fibrous histiocytoma, leiomyosarcoma, synovial sarcoma, solitary fibrous tumor, angioendothelioma, granulosa cell tumor, and others often show CD99 positivity, which assists in diagnosis.
The staining pattern of CD99 also has diagnostic significance; in solid pseudopapillary tumors of the pancreas, its expression is perinuclear dot positive, whereas other neuroendocrine tumors typically show membranous staining.
