The principle of acid-fast staining is based on the fact that the cell walls of acid-fast bacteria such as Mycobacterium tuberculosis contain a large amount of lipids, which can resist decolorization by acid alcohol. After staining with carbol fuchsin, acid-fast bacteria appear red, while other non-acid-fast bacteria and background substances are decolorized by acid alcohol and then counterstained with methylene blue to appear blue. It is mainly used to detect acid-fast bacteria such as Mycobacterium tuberculosis and is an important method for diagnosing tuberculosis and other acid-fast bacterial infections.
