MART-1 is a cytotoxic T lymphocyte-recognized melanocyte antigen, a transmembrane protein expressed in most melanocytes. It is present in melanocytes, adrenal cortex, sex cord stromal cells (strong), and granular cells. Its main application is in the diagnosis and differential diagnosis of malignant melanoma. Spindle cell melanoma shows 50% positive expression, with sensitivity superior to HMB45. MART-1 is positive in adrenal cortical tumors (weakly positive, inconsistent), sustentacular-stromal cell tumors, and granular cell tumors. It is also positive in angiomyolipoma, lymphangioleiomyomatosis, perivascular epithelioid cell tumor, and clear cell sarcoma.
