The principle of Giemsa staining is based on the differential affinity of dyes for various components within cells. It consists of methylene blue and eosin; methylene blue has a strong affinity for DNA and RNA in the cell nucleus, while eosin mainly binds to cytoplasmic components. It is primarily used for blood and bone marrow smear examinations, enabling clear differentiation of various blood cells, such as distinguishing lymphocytes, granulocytes, and other types of white blood cells, aiding in the diagnosis of hematological diseases. It is also used for chromosome analysis to help observe chromosome morphology and number.
