S100 is a soluble acidic protein, composed of α and β chains forming a dimer, capable of existing in three structural forms: S100ao (α chain), S100a (α chain-β chain), and S100b (β chain). It is widely present in mesenchymal-derived cells and lymphohematopoietic tissues, such as glial cells, melanocytes, chondrocytes, Schwann cells, adipocytes, myoepithelial cells, follicular dendritic cells, and Langerhans cells. It is mainly expressed in nervous tissue, pituitary gland, carotid body, adrenal medulla, salivary glands, and some mesenchymal tissues. It is primarily used for the diagnosis and differential diagnosis of astrocytoma, glioblastoma, oligodendroglioma, ependymoma, schwannoma, neurofibroma, malignant melanoma, and liposarcoma. S100 has high sensitivity for all types of malignant melanoma and is often used in combination with Melan A and Melanoma for melanoma diagnosis. It can be used for differential diagnosis of the following tumors: (1) Liposarcoma versus other myxoid tumors; (2) Schwannoma versus leiomyoma; (3) Sclerosing adenosis of the breast versus tubular adenocarcinoma of the breast; (4) Spindle cell type malignant melanoma versus spindle cell carcinoma.
