High molecular weight keratin, mainly reacts with CK1, CK5, CK10, CK14; marks squamous epithelium, ductal epithelium, stratified epithelium, prostate basal cells, and breast myoepithelial cells.
Applications:
1. Marks prostate basal cells to differentiate prostate cancer; distinguishes high-grade PIN from prostate adenocarcinoma; combined use with P63 and AMACR offers better specificity and sensitivity than using 34βE12 alone;
2. Differentiates urothelial atypical hyperplasia (marks only the basal layer) from carcinoma in situ (all epithelial layers positive).
