MyoD1 (myogenic differentiation 1) is a phosphorylated protein with a molecular weight of 45 kDa and serves as a specific marker for rhabdomyosarcoma. It is expressed in embryonic skeletal muscle cells but not in normal adult skeletal muscle cells. It primarily marks small cell and primitive tumor cells, whereas Myogenin mainly marks dedifferentiated tumor cells. It is mainly used for the diagnosis of rhabdomyosarcoma. In the diagnostic application of rhabdomyosarcoma, compared to the common cytoplasmic staining seen with MyoD1 (5.8A), MyoD1 (MX049) has higher sensitivity and nuclear specificity, which can avoid interference in diagnosis caused by cytoplasmic staining.
