PCNA (Proliferating Cell Nuclear Antigen) is a 36kDa nuclear protein associated with the cell cycle and is essential for cellular DNA synthesis. There are two types of PCNA in the nucleus: soluble and insoluble. Soluble PCNA is expressed throughout all phases of the cell cycle, with its amount remaining relatively constant during DNA synthesis, but it is easily extracted by detergents and destroyed by methanol. Insoluble PCNA is more stable; it is not significantly expressed in G0 phase cells, gradually increases during G1 phase, peaks in S phase, and decreases markedly during G2 to M phases. The changes in its amount correspond with DNA synthesis. PCNA can serve as a marker to evaluate the cell proliferation index, used to study the proliferation status of tumor cells, and has certain significance for tumor treatment and prognosis assessment.
