The SOX (SRY-related high-mobility group box) family is a group of nuclear transcription factors related to early embryonic development, involved in processes such as carcinogenic transformation and cancer cell stemness. Based on the homology of their domain sequences, they are divided into 8 subfamilies. SOX-9 belongs to the E subfamily of the SOX family and plays a key role in the development of important organs and tissues such as the nervous system, skeleton, cardiovascular system, and reproductive system. For example, it has an important regulatory role in testis development and estrogen receptor expression during sex determination; in cartilage development, it participates in chondrocyte differentiation and matrix synthesis. Studies have confirmed that high expression of SOX-9 is positively correlated with tumor occurrence and progression, and in many advanced cancers, increased SOX-9 expression levels are observed along with a decrease in overall survival rates of tumor patients.
