NKX2.2 (NK2 Homeobox 2) is a member of the NK2 family of transcription factors, and the gene encoding the NKX2.2 protein is located on chromosome 20. NKX2.2 plays a crucial role in the development and differentiation of the central nervous system, gastrointestinal tract, neuroendocrine cells, and glial cells. It is expressed in normal human tissues such as the brain, pancreatic islet cells, pituitary gland, and gastrointestinal tract. In small round cell tumors, it is highly expressed in Ewing sarcoma, whereas it is lowly expressed in non-Ewing small round cell tumors, making it a specific marker for Ewing sarcoma.
