FOXL2 is a 2.7kb single-exon gene located at the 3q23 region of chromosome 3, encoding 376 amino acids, and belongs to the FOXL family. The normal expression of the FOXL2 gene is an extremely important fundamental condition for maintaining female sexual characteristics. In addition, mutations in the FOXL2 gene are associated with premature ovarian failure, and FOXL2 is considered a regulatory factor in the early differentiation of the ovary. The FOXL2 (402C→G) mutation is present in ovarian sex cord-stromal tumors, but this mutation is absent in various epithelial malignant tumors. Therefore, FOXL2 is a relatively sensitive (e.g., 97.6% positivity rate in granulosa cell tumors) and highly specific marker for diagnosing ovarian sex cord-stromal tumors. When used in combination with inhibin and calretinin, it can improve the accuracy of SCST diagnosis.
