H3.3 G34W, also known as H3F3A G34W, is a mutation occurring at the G34 site of the H3F3A gene on chromosome 1. The mutation types include G34W/R/V/L, among which G34W is the most common. Tumors such as giant cell tumor of bone, bone cyst, chondroblastoma, and osteosarcoma share morphological similarities, making differential diagnosis of these tumors very challenging. H3F3A G34 mutations are present in 85-96% of giant cell tumors of bone; H3.3 G34W is positively expressed in 85-92% of giant cell tumors of bone and is absent in other diseases with similar histological structures. It can serve as a specific marker for diagnosing giant cell tumor of bone.
