GATA-3 belongs to the GATA transcription factor family (including GATA1-GATA6) and is a highly specific marker for breast cancer and bladder cancer, showing negative expression in prostate cancer and kidney cancer. GATA3 is expressed in the brain, parathyroid gland, skin, inner ear, adrenal gland, placenta, embryonic liver, early erythrocytes, and early T cells during embryonic development. From a histological perspective, GATA3 is expressed in adult T lymphocytes, breast glandular epithelial cells, parathyroid gland, and urothelium.
The positivity rate of GATA3 in urothelial carcinoma is 80%-100%, in breast cancer it is 47%-100% (lower in triple-negative breast cancer, higher in glandular breast cancer, over 90%), and it is also high in parathyroid carcinoma.
Additionally, GATA3 can be expressed in salivary duct carcinoma, Hodgkin lymphoma, and paraganglioma. It is generally used in combination with P63 and S100P, two markers of urothelial carcinoma, for differential diagnosis of tumors originating from urothelial cells and tumors of unknown origin.
GATA3 is positively expressed in all non-tumorous trophoblastic proliferative tissues and 81% of trophoblastic tumor tissues, which can be used to differentiate trophoblastic tumors from synovial sarcoma (which does not express GATA3) and squamous cell carcinoma (3/126 cases).
