PSA (Prostate Specific Antigen) is a single-chain glycoprotein synthesized by prostate epithelial cells with serine protease activity, having a molecular weight of 34 kDa. It has a very high specificity for prostate gland epithelium and is expressed in normal prostate acinar cells, ductal epithelial cells, and intraluminal secretions. It is also positively expressed in benign prostatic hyperplasia, proliferation, and tumors of the prostate. Combined detection with NKX3.1 is mainly used for the diagnosis of prostate cancer and metastatic prostate cancer, as well as the differential diagnosis between prostate-derived metastatic cancer and non-prostate-derived metastatic cancer; it should also be noted that some high-grade prostate cancers may show negative results. Additionally, positive expression can be observed in tissues such as the urethra, periurethral glands, bladder villous adenocarcinoma, penile Paget's disease, salivary pleomorphic adenoma (carcinoma), breast cancer, renal cell carcinoma, and mature teratoma.
