NKX3.1 (NK3 Homeobox 1) is an androgen-regulated prostate-specific homeobox gene located on chromosome 8p21.2. As a nuclear transcription factor, it plays a crucial role in prostate embryonic development, epithelial differentiation, and tumorigenesis. It acts as a negative regulator of prostate epithelial cell growth. It is highly expressed in benign prostatic hyperplasia and prostate cancer, and can also be expressed in breast cancer and normal testis. It can serve as a specific marker for diagnosing metastatic lesions originating from the prostate and prostate cancer.
