Vimentin is a type III intermediate filament protein with a molecular weight of 57 kDa, abundant and highly conserved. It has complex biological functions, including maintaining cell morphology, preserving cytoplasmic integrity, stabilizing the cytoskeleton, participating in intracellular material transport, and being involved in the complex processes of tumor metastasis as well as the migration, adhesion of tumor cells and endothelial cells, epithelial-mesenchymal transition (EMT) of epithelial carcinoma, and tumor cell apoptosis. It is expressed in various types of mesenchymal cells such as endothelial cells, fibroblasts, dendritic cells, Langerhans cells, and vascular smooth muscle cells, and also in mesodermal epithelial cells such as those in the kidney, endometrium, ovarian surface epithelium, mammary gland, myoepithelial cells of salivary and sweat glands, and thyroid epithelial cells. Due to its broad expression profile and relatively low specificity, its application in tumor diagnosis is limited. However, it can serve as a good positive control marker, easily expressed in non-tumorous epithelial tissue cells, and can be used as an internal control; its expression in formalin-fixed paraffin-embedded stromal tissues can help observe antigen loss caused by over-fixation or improper fixation. It can be used for the diagnosis of mesenchymal-origin tumors and also for the differential diagnosis between endometrial carcinoma and cervical adenocarcinoma.
