PR (Progesterone receptor) is a ligand-activated nuclear transcription regulator and a member of the steroid receptor superfamily. It includes two isoforms, PRA (94kDa) and PRB (114kDa), where PRB is the product of the full gene transcription, and PRA lacks the first 164 amino acids at the amino terminus. PR is a protein with dual functions: it can act as a ligand-dependent transcription factor directly interacting with nuclear DNA to regulate transcription, and it can also regulate extranuclear signaling pathways as a non-transcription factor. It plays a key role in mediating and regulating the functions and reproductive activities of the ovary, uterus, and breast. It is expressed in normal and benign or malignant endometrial and breast epithelial cells. Studies have shown that most tumors positive for ER and PR respond well to endocrine therapy, with high remission rates, low recurrence rates, and good prognosis. Even patients positive for only one of ER or PR have a better prognosis than those negative for both. PR is commonly tested alongside ER and HER-2 in routine examinations of breast cancer patients.
