The NUT (Nuclear protein in testis) gene is located at chromosome 15q14. The NUT protein does not contain a folded three-dimensional structure, but it includes a nuclear export sequence and a nuclear localization sequence, allowing it to shuttle between the cytoplasm and the nucleus. NUT is expressed in germ cells of the testis and ovary. The expression rate of NUT varies among different germ cell tumors, such as seminoma (64-93%), embryonal carcinoma (0-9%), immature teratoma (75%), and spermatocytic tumor (6-74%). Rearrangement of the NUT gene can lead to the development of NUT midline carcinoma (NMC), which mostly occurs in midline organs above the diaphragm, and has also been found in organs below the diaphragm such as the bladder, submandibular gland, parotid gland, and abdominal organs. NMC is an aggressive and highly lethal cancer with an average survival time of less than one year. NUT is primarily used for the diagnosis of NMC. It can also be used for the diagnosis of CIC-NUTM1 sarcoma and germ cell tumors.
