p16 (p16(INK4a)), also known as MTS1 or CDKN2, is an important tumor suppressor gene located on chromosome 9p21. It is involved in the regulation of the cell cycle, negatively regulating cell proliferation and division. It can specifically bind to CDK4, inhibiting CDK4 activity, thereby preventing the cell from progressing from the G1 phase to the S phase. It plays a negative regulatory role in the cell proliferation cycle controlled by the Rb gene. It can serve as a marker for cervical lesions, showing high expression in high-grade cervical intraepithelial neoplasia and tumors infected with high-risk HPV. Positive expression is also observed in cervical cancer, lung cancer, malignant melanoma, breast cancer, ovarian cancer, liposarcoma, and other tumors. It can be used for the diagnosis of cervical adenocarcinoma and endometrial cancer and is of great significance for the detection of cervical cancer.
