PTEN (Phosphatase and tensin homolog deleted on chromosome 10), also known as MMAC1 or TEP1, is located at chromosome 10q23.3. It is a tumor suppressor gene and a major negative regulator of the PI3K/Akt pathway. The PTEN protein has protein phosphatase and lipid phosphatase activities and plays an important role in cell growth, apoptosis, adhesion, migration, and invasion. This gene exhibits heterozygous loss or mutation in many tumors, including gliomas, ovarian cancer, endometrial cancer, prostate cancer, melanoma, liver cancer, and kidney cancer. It is mainly used in research on breast cancer, malignant melanoma, endometrial cancer, bladder cancer, small cell lung cancer, colorectal cancer, and ovarian endometrioid carcinoma. Compared with uterine serous carcinoma, high-grade endometrioid carcinoma often shows loss of PTEN expression, which can be used for differential diagnosis between uterine serous carcinoma and endometrioid carcinoma. In addition, loss of PTEN expression is also commonly seen in Cowden syndrome.
