Pax-8 (Paired box gene 8) is a member of the Pax nuclear transcription factor family, located on chromosome 2q13. It is involved in the development of thyroid follicular cells and the expression of thyroid-specific genes. It is expressed in normal thyroid tissue and related tumors, non-ciliated mucous cells of the fallopian tube, and simple ovarian cysts, but not in normal ovarian surface epithelial cells. It is highly expressed in ovarian serous carcinoma, while almost not expressed in ovarian mucinous carcinoma; its expression profile in kidney tumors is similar to Pax-2, with a higher expression rate than the latter; it is not expressed in normal lung tissue, lung cancer, or breast cancer. Combined with organ system-specific markers (such as Uroplakin III, Mammaglobin, and TTF-1), it is used to identify the primary site of metastatic cancers of unknown origin or to differentiate certain epithelial tumors. It can serve as a highly sensitive and specific marker for renal cell carcinoma and non-mucinous ovarian carcinoma.
