Calponin is an actin-binding protein that inhibits smooth muscle cell contraction. It is positively expressed in normal smooth muscle cells and myoepithelial cells, and myofibroblasts in connective tissue hyperplasia stroma also show positive expression. It is mainly used for the diagnosis of smooth muscle tumors and myoepithelial cell tumors, and can also be used in the diagnosis of tissue lesions containing myoepithelial cells such as the breast, to detect the loss or distribution of myoepithelium. It helps differentiate between invasive ductal carcinoma and ductal carcinoma in situ of the breast.
Calponin is positive in vascular leiomyoma, neurofibromyxoma, hemangioma, bone myofibrosarcoma, and some focal synovial sarcomas.
