TdT (Terminal deoxynucleotidyl transferase) is a DNA polymerase with a molecular weight of 58.3 kDa, located at chromosome 10q23-q24. It is expressed in most cortical thymocytes, pro-T cells, pro-B cells, thymus, and multipotent stem cells in the bone marrow. Tumors derived from immature lymphocytes show increased TdT activity; similar cases are observed in lymphoblastic leukemia, acute T/B lymphoblastic leukemia, etc. Positive expression can also be seen in some Merkel cell carcinomas and blastic plasmacytoid dendritic cell tumors, while other mature (peripheral) malignant lymphomas are negative. It is an important marker for studying lymphomas and leukemias. It can be used for the differential diagnosis between lymphocytic lymphoma and Burkitt lymphoma (Note: Burkitt lymphoma may sometimes show very weak expression).
