SATB2 (Special AT-rich sequence-binding protein 2) is located at chromosome 2q32-q33, composed of 733 amino acids with a molecular weight of 83 kDa, containing one homologous domain, and exhibits significant evolutionary conservation. It can specifically bind to DNA nuclear matrix attachment regions and plays an important role in regulating chromatin transcription and remodeling, cortical neuron differentiation, and osteoblast development. It shows tissue specificity, being almost not expressed in normal human tissues, but selectively expressed in epithelial-derived tissues and glandular cells of the lower digestive tract. It is positively expressed in benign and malignant bone tumors containing osteoblast differentiation and soft tissue tumors with heterotopic bone differentiation, serving as a specific marker for osteoblast differentiation. It is mainly used for the diagnosis of osteosarcoma and differential diagnosis from other non-osteogenic sarcomas. Additionally, it can serve as a sensitive and specific marker for lower digestive tract tumors (such as colorectal cancer) when combined with CK7 and CK20. It is highly expressed in colorectal cancer, while other tumors such as pancreatic, gastric, gallbladder, ovarian, cervical, and endometrial adenocarcinomas rarely express it or are negative.
