TOPO II a (Topoisomerase II alpha) is a nuclear matrix enzyme protein with a molecular weight of 170 kDa, located at chromosome 17q21.3. It has phosphodiesterase activity and can form stable covalent complexes with DNA. Its levels increase during the cell proliferation phase. In somatic cell nuclei, it regulates the dynamic changes of nucleic acid spatial conformation and plays a key role in DNA unwinding, breakage, and the ligation and repositioning of DNA ends after replication. The expression level of TOPO IIa is closely related to the prognosis of tumor patients and directly affects the choice of chemotherapy drugs. A decrease in expression levels leads to resistance to TOPO II inhibitor drugs. It is commonly used as an indicator for assessing multidrug resistance in tumors and as a target for anticancer drug research.
