SSTR2 (Somatostatin receptor 2) is a G-protein-coupled cell membrane receptor. It is widely expressed in various normal organs and tissues, such as the pancreas, brain, kidneys, jejunum, colon, and liver. In tumors, it is mainly expressed in various neuroendocrine tumors and multiple solid tumors, such as pancreatic neuroendocrine tumors, pituitary adenomas, meningiomas, breast cancer, small cell lung cancer, and hormone-producing gastrointestinal tumors. Additionally, it is generally positively expressed in normal prostate tissue, whereas most prostate cancers are negative. Data indicate that SSTR2 can serve as a prognostic marker for octreotide drugs. Peptide receptor radionuclide therapy (PRRT) is one of the treatment methods for patients with metastatic neuroendocrine tumors; if the tumor tissue is negative for SSTR2 expression, radionuclide targeted therapy cannot be used. It is an independent predictor for improved overall survival and progression-free survival in neuroendocrine tumors; its expression in medullary thyroid carcinoma is associated with increased survival rates; and it is related to poor prognosis in hepatocellular carcinoma.
