S100P is a member of the S100 protein family, located on chromosome 4p16, with a molecular weight of 10.4 kDa, and features the typical two EF-hand structures of S100 proteins. It is involved in tumor cell growth, proliferation, invasion, metastasis, and angiogenesis. It was first found to be highly expressed in placental tissue and is widely distributed in normal human tissues such as the heart, brain, lungs, bone marrow, placenta, and liver. It shows positive expression in colorectal cancer, breast cancer, prostate cancer, and non-small cell lung cancer; is highly expressed in pancreatic ductal adenocarcinoma, while benign pancreatic ducts and acinar glands do not express it. It has been reported that S100P is expressed in the invasive components of all intraductal papillary mucinous tumors, including nerves, lymph nodes, and microinvasion. 90% of primary cholangiocarcinoma biopsies are strongly positive for S100P, whereas benign biopsy tissues are negative. Therefore, S100P can be used to distinguish cancer from reactive epithelial changes in cholangiocarcinoma biopsy specimens. It is also one of the specific markers for urothelial carcinoma.
