STAT6 (Signal transducers and activators of transcription 6) is a member of the STAT protein family. It is involved not only in the activation of the IL-4 signaling pathway and regulation of the immune system but also in cell proliferation and apoptosis. Solitary fibrous tumors (SFT) can occur in various locations and are mostly benign, with about 10% of cases showing aggressive behavior. Although most SFTs can be diagnosed based on histological features and CD34 expression, their diversity sometimes makes diagnosis challenging. Studies have shown that most benign and malignant SFTs harbor a specific NAB2-STAT6 fusion gene. The fusion of NAB2 and STAT6 genes causes STAT6 in the cytoplasm to translocate into the nucleus, leading to strong nuclear expression of STAT6. Therefore, STAT6 can serve as a highly sensitive and specific marker for SFT, aiding in the diagnosis of SFT and differential diagnosis from similar lesions. Additionally, STAT6 is highly expressed in various malignant tumors and can be used as a prognostic indicator.
