TFE3 (Transcription factor E3) is a member of the MiT transcription factor family, located on chromosome Xp11.2. It has many important biological functions, such as involvement in cell growth and maintenance, DNA-dependent transcription regulation, and tRNA aminoacylation during protein translation. Rearrangements of the TFE3 gene are associated with various tumors, including Xp11.2 translocation/TFE3 gene fusion-related renal cell carcinoma, alveolar soft part sarcoma (ASPS), perivascular epithelioid cell tumor (PEComa), granular cell tumor, and some ossifying fibromyxoid tumors. A characteristic of Xp11.2 translocation/TFE3 gene fusion-related renal cell carcinoma is the occurrence of a translocation at Xp11.2 in the genome, forming a new fusion gene that leads to overexpression of the transcription factor TFE3. ASPS is a rare soft tissue sarcoma of uncertain differentiation, typically occurring in the limbs of young patients, characterized by rearrangements on chromosomes 17q25 and Xp11.2, producing the ASPSCR1-TFE3 fusion gene and abnormal transcription, thereby causing tumor development. TFE3-positive PEComa has distinct clinicopathological features, with younger patients predominantly female, and tumor cells exhibiting epithelioid or spindle shapes. TFE3 protein is expressed in various tissues, especially in the genitourinary tract. It can be used for the diagnosis of Xp11.2 translocation/TFE3 gene fusion-related renal cell carcinoma and ASPS.
